1629 High-throughput T cell receptor sequencing aids in the diagnosis of CTCL

Cutaneous T-cell lymphomas (CTCL) are difficult to diagnose and may resemble benign inflammatory dermatoses (BID). Hence, diagnosis requires consideration of clinical, histopathologic, molecular studies. Though, widely used studies rely on the T cell receptor g (TCRg) polymerase chain reaction (PCR) detect a clone, it is reportedly positive in ∼50% early CTCL cases. High throughput TCR sequencing (HTS) TCRb gene (ImmunoSEQ®) prior cohort has demonstrated more sensitive specific than TCRg PCR discriminates from BID. We evaluated performance clonoSEQ®, clinically available assay for detection dominate clone by HTS patients with suspected referred multi-disciplinary cutaneous lymphoma clinic at DFCI 2016-2021.
